Oncology FISH Assays

Available Fluorescence in situ Hybridization Assays

Specimen Requirements
5mL (3mL) Whole blood or Bone marrow in a Dark Green Sodium Heparin tube. Keep specimen at room temperature.  Note: If a Bone Core Biopsy (not the liquid aspirate) is being sent for Chromosomes and or FISH study, the preferred media is RPMI, as this is a nutrient, and it keeps the sample moist and prevents desiccation. Refer to the test name below in the Test Directory for more instructions:

AML, AML1/ETO Translocation 8,21 by FISH:

[t(8;21)(q22;q22)]
Order Name: AML 8-21T
Test Number: 0116200
Clinical Use

Frequently observed karyotic abnormality associated with Acute Myeloid Leukemia (AML), especially FAB M2.

AML, CBFB/MYH11, Inversion 16 by FISH:

[inv(16) or t(16;16)(p13;q22)]
Order Name: AML INV16
Test Number: 0116175
Clinical Use
inv(16), AML-M4 CBFB/MYH11 is associated with acute myeloid leukemia (FAB M4 Eo subtype)

AML, 11q23 Gene Rearrangement by FISH:

(11Q23)
Order Name: AML 11Q23
Test Number: 0116225
Clinical Use
11q23 band (11q23+) bearing the MLL gene translocation (MLL+) is a recurrent chromosome change observed in 3% to 7% of acute lymphoblastic leukemias and in 3% to 4% of acute myeloblastic leukemias.

AML, M3, PML/RARA by FISH:

[ t(15;17)]
Order Name: AML M3 FSH
Test Number: 0116025
Clinical Use
Useful for diagnosing or excluding acute promyelocytic leukemia (AML M3) with the standard translocation. It will not detect variant translocations seen in AML M3, such as the t(11;17) or t(5;17).

BCR/ABL (Philadelphia Chromosome) Qualitative by FISH:

CML/ALL, bcr/abl, [t( 9,22)]
Order Name: PHILA CHRO
Test Number: 0113275
Clinical Use
Useful for diagnosing chronic myelogenous leukemia (CML), following course of treatment in CML, and as a prognostic marker in B-cell acute lymphoblastic leukemia (ALL). In addition, it is useful in excluding CML if other myeloproliferative disorders are in the differential diagnosis.

B-Cell Chronic Lymphocytic Leukemia by FISH:

[11qA/P, +12, 13q-, 17p-, 6q23 (MYB)]
Order Name: B CELL CLL
Test Number: 0115660
Clinical Use
Useful for providing prognostic information in known diagnoses of B-cell chronic lymphocytic leukemia/small lymphocytic lymphoma (CLL/SLL) and in multiple myeloma cases.

Burkitt's Lymphoma/NHL/ALL, IGH/MYC by FISH:

[t(8;14)]
Order Name: BURKI FISH
Test Number: 0117900
Clinical Use
Useful to diagnose Burkitt's type lymphoma and some cases of acute lymphoblastic leukemia.

Follicular Lymphoma, IGH/BCL2, by FISH:

[t(14;18)(q32;18q21.3)]
Order Name: FOLLC FISH
Test Number: 0116975
Clinical Use
Useful to diagnose follicular lymphoma (&gt75%) and some diffuse large B-cell lymphomas.

Mantle Cell Lymphoma, IGH/CCND1 by FISH:

[t(11;14)]
Order Name: MANTL FISH
Test Number: 0116800
Clinical Use
Useful to detect classical translocation in Mantle cell lymphoma.

Multiple Myeloma Profile by FISH:

[13q-, 17p-, rea 14q32]
Order Name: MULT MYELO
Test Number: 0115525
Clinical Use
Multiple Myeloma (MM) is characterized by the proliferation of malignant monoclonal plasma cells in the bone marrow. In most cases, there is low proliferation index of terminally different malignant plasma cells. As a result, conventional cytogenetic analysis frequently yields normal results. The most frequent abnormalities with prognostic association have been included in this panel (13q-, 14q rearrangements and 17p-). Interphase FISH studies can enhance the detection rate and complement conventional cytogenetic techniques.

Myeloid Disorders Profile by FISH:

[-5/5q-, -7/7q-, +8, 20q-]
Order Name: MYELOID PR
Test Number: 0115525
Clinical Use
Useful for diagnosing some types of myelodysplastic syndromes with certain specific cytogenetic abnormalities (MDS) as well as certain subtypes of MDS such as 5q- syndrome. It is also useful as adjunct test in diagnosing chronic myelomonocytic leukemia (CMML) and juvenile myelomonocytic leukemia (JMML) and for following the evolution of chronic myelogenous leukemia (CML) to accelerated phase or blast crisis.
